Metatropic dysplasia is caused by pathogenic variants in the TRPV4 gene. The TRPV4 protein plays a role in regulating calcium ion channels that are important for skeletal development. As this is an autosomal dominant condition, a single altered copy of the gene is sufficient to cause the disorder. Affected individuals have a 50% chance of passing the variant to their offspring, and genetic counseling is recommended to help families understand these implications.
